The dose-response question is the one we get most. Should you take 250 mg, 500 mg, 1,000 mg, more? Almost every NMN brand sells multiple strengths and offers conflicting advice, which doesn't help. The short answer is that the human evidence forms a fairly clear picture, one that most marketing flattens. This piece sets out what the trials show across doses, where the returns start to diminish, and how to think about your own dose whether you're starting NMN or already taking it.

What the dose-response trials show
Across roughly a dozen randomised controlled trials in humans, NMN has been studied at doses ranging from 100 mg to 1,250 mg per day, mostly given orally and mostly for 8 to 12 weeks. The pattern that emerges is consistent. Doses below 250 mg raise NAD+, but only modestly. Doses between 250 mg and 600 mg produce a clean dose-response, with 500 mg showing the largest reproducible effect on functional outcomes. Doses above 600 mg continue to raise blood NAD+, but the additional effect on tissue function and reported outcomes is small.
The most useful single dataset is the Pencina et al. 2023 trial, which compared 300 mg, 600 mg, and 900 mg of NMN against placebo for 60 days in overweight and obese middle-aged and older adults. NAD+ rose at all three doses, but the relationship was non-linear. The jump from 300 to 600 mg added meaningful blood NAD+ and improved walking endurance. The jump from 600 to 900 mg added blood NAD+ but produced little additional functional benefit. That ceiling is consistent with a transporter-saturation effect at the cell membrane, which has been observed in animal pharmacokinetic studies.
The 250 mg case
For most adults under 50 with no specific health concern, 250 mg is the entry-level dose where benefit becomes reliable. The case for it has three parts.
First, the NAD+ deficit at 30 to 50 is real but moderate compared with the deficit at 60-plus. You're not trying to fully overcome a 60 percent decline; you're nudging a 25 to 35 percent decline back toward baseline, and the dose to do that doesn't need to be heroic.
Second, 250 mg is enough to push intracellular NAD+ measurably higher in most healthy adults. In AVEA's customer survey of 280 NMN users, 80 percent reported better energy and focus within 30 days.
Third, 250 mg is the most cost-efficient dose. NMN is not a cheap molecule, and at 250 mg per day you're paying half what you would for 500 mg, often without halving the benefit. For preventive use in someone whose lifestyle is already supportive, the maths favours 250 mg.
The 500 mg case
500 mg is where the human trial data is strongest, for older adults, people with existing fatigue or recovery issues, and anyone who has tried 250 mg without noticing much.
The evidence around it brackets the dose. In postmenopausal women with prediabetes, 250 mg per day produced measurable insulin sensitivity gains (Yoshino et al., 2021), and the largest functional improvements in Pencina's dose-response trial came at 600 mg (Pencina et al., 2023). Trials in older adults have reported gains in walking distance, muscle function, and fatigue at doses in this range.
For most people over 50, or anyone whose primary goal is recovery, exercise capacity, or reversing a deeper NAD+ deficit, 500 mg is the dose with the most evidence behind it, which is why AVEA's NMN is available in a 500 mg "Forte" formulation for this group. The functional improvements at 500 mg tend to show up faster, often by week 3 or 4, and are more consistent across responders. The trade-off is cost: if you're young and metabolically healthy, the extra 250 mg may be over-spending.
Doses above 600 mg: where evidence ends
Some longevity protocols use 1,000 mg or more, often citing higher doses as "more aggressive" anti-ageing support. The evidence does not currently support this.
Pharmacokinetic studies suggest that the cellular transporters that move NMN from blood into cells become saturated above roughly 500 mg in a single dose. Above that point, additional NMN circulates in the blood without entering cells efficiently, much of it gets converted to nicotinamide and excreted, and the dose-response curve flattens.
In Pencina et al. 2023, the jump from 600 mg to 900 mg added blood NAD+ but produced little additional functional benefit. No published trial has shown a clear incremental advantage at doses above 600 mg over a 500 mg comparison.
There's a separate question about safety at higher doses. NMN has been well tolerated across the doses studied, including a trial that went up to 1,250 mg per day, with no major adverse signals (Igarashi et al., 2022). Long-term safety data above 1 g per day is more limited. For most people, the better strategy is "right dose for the goal" rather than "more is better."
A possible exception is split dosing: 500 mg in the morning and 500 mg with lunch could, in theory, maintain higher steady-state NAD+ than a single 1,000 mg morning dose, by feeding the transporters across two windows rather than one. This is mechanistically plausible but lacks direct trial support.
Body weight, age, and individual factors
NMN trials don't typically dose by body weight, but a few real-world factors influence where you should sit on the dose curve.
Age matters most. The NAMPT bottleneck, CD38 expansion, and accumulated DNA damage are all stronger drivers of NAD+ depletion in your 60s than in your 40s. If you're over 60, default toward 500 mg; if you're under 40, 250 mg is usually enough.
Body weight matters less than people assume, but at the extremes it plays a role. A 100 kg adult with high muscle mass distributes the same oral dose across more tissue than a 55 kg adult, and may need to lean toward the higher end of the range for an equivalent intracellular effect. This is anecdotal, since no trial has formally weight-dosed NMN, but it's a reasonable adjustment.
Goal matters. If you're using NMN as preventive insurance, lower doses are appropriate. If you're addressing a specific issue (recovery in masters athletes, post-illness fatigue, persistent cognitive sluggishness), the higher end of the evidence-supported range is easier to justify.
Methylation status is a factor people forget. NMN is metabolised through nicotinamide, which consumes methyl groups during clearance. Some people benefit from pairing higher NMN doses with TMG (trimethylglycine), or from keeping folate and B12 status adequate, to avoid hidden methylation pressure.
How to decide if you should increase
The most practical approach is to start at 250 mg, run it for 8 to 12 weeks, and assess. If the energy, sleep, and recovery markers you care about are moving in the right direction, stay where you are. If you're not noticing change after 8 weeks on a quality NMN, step up to 500 mg before concluding NMN doesn't work for you.
